{"id":357,"date":"2018-12-20T16:40:51","date_gmt":"2018-12-20T16:40:51","guid":{"rendered":"http:\/\/radparts.com\/cpswp\/?p=357"},"modified":"2018-12-20T16:40:51","modified_gmt":"2018-12-20T16:40:51","slug":"trial-results-show-compound-makes-radiotherapy-more-effective","status":"publish","type":"post","link":"https:\/\/radparts.com\/cpswp\/uncategorized\/trial-results-show-compound-makes-radiotherapy-more-effective\/","title":{"rendered":"Trial results show compound makes radiotherapy more effective"},"content":{"rendered":"\n<p>A trial has shown that radiotherapy is more effective when levels of ropidoxuridine in a patients\u2019 body reach a certain level\u2026<\/p>\n\n\n\n<figure class=\"wp-block-image\"><img decoding=\"async\" src=\"https:\/\/dlg7f0e93aole.cloudfront.net\/wp-content\/uploads\/radiotherapy-1.jpg\" alt=\"radiotherapy\"\/><\/figure>\n\n\n\n<p>A\n new drug designed to make radiotherapy more effective in treating \ncancer has been given to patients while they are receiving radiation and\n shown to be safe.<\/p>\n\n\n\n<p>The drug, called \n5-iodo-2-pyrimidinone-2\u2032-deoxribose (IPdR), or ropidoxuridine, has the \nadvantage that patients can take it in capsule form, as opposed to \nintravenously. When the drug enters the body, researchers believe it \nchanges into an active form that can make cancer cells more susceptible \nto the effects of radiotherapy.<\/p>\n\n\n\n<p>Results of US NCI trial #9882, presented by Dr Timothy Kinsella from the Department of Radiation Oncology at the Warren Alpert Medical School of Brown University and Rhode Island Hospital  in the USA, show that the drug has minimal side effects when given to  patients with a variety of gastrointestinal cancers during the course of  their radiotherapy.<\/p>\n\n\n\n<p><strong>Webinar: Improve ultrapure water system performance and uptime with ozone<\/strong><\/p>\n\n\n\n<p>Register for our on-demand webinar with \nSUEZ which is designed to help companies understand the application and \nbenefit of ozone technology in ultrapure water systems and how ozone \nsolutions can improve water system performance, uptime and product \nquality in pharmaceutical manufacturing applications.<\/p>\n\n\n\n<p>Dr Kinsella explained: \u201cThe aim of my research is to find  better ways to treat patients with cancer, and specifically to develop  ways to make <a href=\"https:\/\/radparts.com\">radiation treatment <\/a>safer and more effective.<\/p>\n\n\n\n<p>\u201cPrevious\n research found a promising compound called iododeoxyuridine, or IUdR, \nthat worked very well to improve the effectiveness of radiotherapy, but \nIUdR could only be given intravenously and proved to have many side \neffects for patients.<\/p>\n\n\n\n<p>\u201cAs a result, this new drug, IPdR, was \ndeveloped. It\u2019s a prodrug that can be taken as a capsule and, once \ninside the body, it\u2019s converted into the active drug, IUdR.<\/p>\n\n\n\n<p>\u201cThis \ntrial is the first to test it out in patients while they are receiving \nradiation therapy, and the results suggest that it\u2019s safe with minimal \nside effects.\u201d<\/p>\n\n\n\n<p>Dr Kinsella and his colleagues tested the new drug \nin a group of 18 patients with advanced cancers including oesophageal, \npancreatic, liver, bile duct, rectal and anal cancers. All had been \nreferred for palliative radiotherapy.<\/p>\n\n\n\n<p>Alongside their \nradiotherapy, patients were given a daily dose of the IPdR prodrug over \n28 days. They were given blood tests to check on the levels of both the \nIPdR prodrug and the active IUdR drug at various points during their \ntreatment. The dose of the prodrug was gradually increased, and patients\n were monitored for side effects.<\/p>\n\n\n\n<p>Results of the trial suggest \nthat IPdR can be safely given to patients up to a dose of 1200mg per day\n for 28 days without causing serious side effects. The results also \nsuggest that this dose creates levels of the active IUdR drug in \npatients\u2019 blood that are high enough to have a radiosensitising effect.<\/p>\n\n\n\n<p>Of\n the 18 patients on the trial, 14 could be assessed for any effect on \ntheir tumours with a CT or MRI scan 54 days after beginning the \ntreatment. Among these patients, one had a complete response \n(disappearance of tumour), three showed a partial response (at least 30 \npercent reduction in the tumour targeted by radiotherapy), nine had \nstable diseases (no growth in the tumour) and one patient stopped \ntreatment because of an infection and had progressive disease (at least \n20 percent growth in the tumour).<\/p>\n\n\n\n<p>Dr Kinsella added: \u201cThis \nclinical trial showed that when patients take IPdR at home before coming\n for radiation treatment, the level of IUdR in their bloodstream is high\n enough to make radiation more effective at killing cancer cells. It \nalso showed that the dose of IPdR needed to achieve therapeutic levels \nof IUdR in the blood causes minimal side effects.<\/p>\n\n\n\n<p>\u201cHowever, this \ntrial was with patients who had recurrent cancer and had already \nreceived a number of other cancer treatments. In newly diagnosed \npatients, it could be that we can safely use a higher dose and have a \nbigger effect on tumours.\u201d<\/p>\n\n\n\n<p>Dr Kinsella and his colleagues are \nalready studying the effects of IPdR in patients receiving whole brain \nradiotherapy for cancer that has spread to the brain. Following this \ntrial, plans are in progress to study the drug in patients who have been\n newly diagnosed with glioblastoma, an aggressive form of brain cancer.<\/p>\n\n\n\n<p>Dr Eric Deutsch, Professor of Radiation Oncology and head of the radiation oncology department and research unit at the Institut Gustave Roussy, Villejuif,  France, is a member of the EORTC-NCI-AACR Symposium scientific  committee and was not involved with the research. He commented:  \u201cRadiotherapy is a vital element in treating many forms of cancer. This  research is investigating whether the IPdR drug could make radiotherapy  even more effective for more patients.<\/p>\n\n\n\n<p>\u201cIn treating cancer \npatients, we must always consider the risks and benefits of any therapy.\n In this study, the risks of the IPdR drug were minimal, and the benefit\n was that it can be taken by patients at home.
